Elk-1, a 62kD transcription factor, is the most abundant component of ternary complex factors that is regulated by phosphorylation and in turn regulates gene expression in response to multiple stimuli. Elk-1 is a substrate of MAP kinase and phosphorylated by ERK2 on several sites in the carboxyl terminus, including serine 383. Phosphorylation of Elk-1 on serine 383 is critical for its transcriptional activity, including Elk-1-mediated transcription of c-Fos. Dephosphorylation of serine 383 by calcineurin downregulates Elk-1 transcriptional activity. Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Positive Control: Y-1 adrenocortical tumor cells; NIH3T3 cells +/- EGF Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
