Endophilin 1, Mid, SH3 Domain

Endophilin-1 is a 40kD protein containing a COOH-terminal SH3 domain and an NH2-terminal lipid binding domain1 and it is expressed in the brain. Endophilin dimerises through its N-terminus, and participates at multiple stages in clathrin-coated endocytosis, from early membrane invagination to synaptic vesicle uncoating. Both its C-terminal SH3 domain and N-terminus are required for endocytosis. Through its SH3 domain, endophilin bound to proline-rich domains (PRDs) in other endocytic proteins, including synaptojanin and dynamin. The N-terminal region possesses unique functions affecting lipid membrane curvature, through\nlysophosphatidic acid acyl transferase (LPAAT) activity and direct binding and tubulating activity. In addition to synaptic vesicle formation, endophilin-1 complexes with signalling molecules, including cell surface receptors, metalloprotease disintegrins and germinal centre kinase-like kinase (GLK). Therefore, endophilin-1 may serve to couple vesicle biogenesis with intracellular\nsignalling cascades. Endophilin is implicated in endocytosis and recycling of synaptic vesicles and it was suggested that endophilin-1 works with dynamin to mediate synaptic vesicle invagination from the plasma membrane and fission. Endophilin may also participate in membrane deformation and vesicle budding.\n\nApplications:\nSuitable for use in ELISA and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nELISA: 0.01-1ug/ml\nWestern Blot: 1-3ug/ml\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Reacts with the mid region of mouse, human and rat Endophilin-1 protein. On Western blots, it identifies a single band at ~40kD. Reactivity has been confirmed with mouse brain.
