Endostatin (Collagen alpha-1(XVIII) Chain, COL18A1)

Endostatin is a naturally occurring 20kD polypeptide derived from the C-terminal portion of type XVIII collagen. It functions as an anti-angiogenic cytokine that is expressed in various organs with the highest levels in liver, lung and kidney. Endostatin inhibits angiogenesis by blocking the pro-angiogenic activities of VEGF and FGF-basic. Recombinant human Endostatin is a 20.2kD protein consisting 184aa residues.\n\nApplications:\nSuitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization. Other applications not tested.\n\nRecommended Dilution:\nELISA: To detect hEndostatin by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required.\nWestern Blot: To detect hEndostatin by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hEndostatin is 1.5-3ng/lane, under either reducing or non-reducing conditions.\nImmunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 0.125ug/ml-0.250ug/ml with an overnight incubation at 4 degrees C. An alkaline phosphatase-labeled polymer detection system was used with a red chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Optimal concentrations and conditions may vary.\nNeutralization: To yield one-half the maximal inhibition of the biological activity of Human Endostatin (0.5ug/ml), a concentration of 2ug/ml of this antibody is required.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
