Enterokinase, Light Chain, Ultrapure, Recombinant, Human (Enteropeptidase)

A serine protease which cleaves proteins at the C-terminal side of Lysine after the sequence Asp-Asp-Aps-Aps-Lys; used to cleave recombinant fusion proteins which have the EK recognition sequence Glu4Lys or Asp4Lys. The light chain of enteropeptidase has full enzymatic activity. No other protease activity was detected. Human enteropeptidase binds specifically to STI-agarose. \n\nUnit Definition:\nOne unit of human enteropeptidase will cleave 2mg of thioredoxin/human EGF fusion protein with the Asp-Asp-Aps-Aps-Lys sequence at the joining point in 22 hours at 4 degrees C, in 16 hours at 25 degrees C or in 8 hours at 37 degrees C. \n\nDilution Buffer:\n50mM Tris-HCl, pH 8.0, 500mM sodium chloride, 1mM calcium chloride, 50% glycerol. \n\nAssay Conditions:\n50mM Tris-HCl or sodium phosphate, pH 8.0 with or without calcium chloride. The enzyme is active at a pH range of 6.0-9.0.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
