EphB4 Receptor Tyrosine Kinase

The Eph receptors are the largest family of receptor tyrosine kinases. They can be grouped into two subclasses: the EphA and the EphB receptors. The ligands for the Eph receptors are the ephrins. The EphA receptors bind ephrin-A ligands, which are tethered to the membrane by a GPI linkage, and the EphB receptors bind ephrin-B ligands, which have a membrane spanning region. The Eph receptors are widely expressed during embryonic development. Ephrins and Eph receptors have been implicated in many developmental processes including neuronal network formation, guidance of cell migration, and axonal pathfinding. They remain expressed at lower levels in adult tissues and are upregulated in pathological conditions such as cancer. EphB4 is expressed in epithelial cells and in endothelial cells of embryonic veins, where its signaling mediates cell migration and proliferation via the PI3K pathway. It is upregulated in several types of tumors, including mammary tumors, colon cancer, and endometrial cancer. EphB4 has previously been designated Htk, Myk1, Tyro11 and Mdk2.\n\nApplications:\nSuitable for use in ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilutions:\nWestern Blot: 1-3ug/ml\nImmunoprecipitation: 5ug/IP reaction\nImmunohistochemistry: 10ug/ml. Formalin-fixed, paraffin-embedded tissues require heat induced epitope retrieval (HIER) with citrate buffer, pH 6.0 prior to staining.\nOptimal dilutions to be determined by the researcher.\n\nPositive Controls:\nWestern Blot: EphB4-transfected 293T cells and human MDA-MB-231 breast carcinoma cell lysates\nImmunohistochemistry: Human colon cancer tissues\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
