Epidermal growth factor receptor (EGFR) is a 170kDa transmembrane glycoprotein receptor tyrosine kinase that, when activated by epidermal growth factor EGF), affects cell growth and differentiation in normal and cancer cells (1). Binding of EGF or TGF alpha to EGFR activates tyrosine kinase activity of the receptor (2). The carboxy terminal tyrosine residues on EGFR, Tyr 1068, Tyr 1148, and Tyr 1173, are the major sites of autophosphorylation, which occurs as a result of EGF binding 3). EGFR overexpression is exhibited various cancer, such as glioma, colorectal carcinoma, breast carcinoma, and head and neck carcinoma (4). The phosphorylation level of EGFR is considered one of the important predicators for outcome in non-small cell lung cancer therapy (5). Applications: Suitable for use in Flow Cytometry, Western Blot, Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 1:1000-5000 Immunocytochemistry: 1: 100-250 Flow Cytometry: 1:55 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
