Epidermal Growth Factor Receptor (erbB, EGFR)

Growth factor receptors are involved not only in promoting the proliferation of normal cells\nbut also in the aberrant growth of many types of human tumors. For example, the epidermal growth factor receptor (EGFR) is found to be mutated and/or overexpressed in many common solid human squamous cell carcinomas including breast, brain, bladder, lung, gastric, head & neck, esophagus, cervix, vulva, ovary, and endometrium. Overexpression of the EGFR gene occurs in carcinomas with and without gene amplification. EGFR is a transmembrane glycoprotein that is a member of a family of protein tyrosine kinases crucial in maintaining a normal balance in cell growth and development. EGFR and erbB-2 are particularly important in\nbreast cancer because increased production or activation has been associated with poor prognosis. Tumor aggressiveness and patient outcome can be directly correlated to their production levels. Inhibitors of EGFR represent an emerging new class of anti-cancer drugs that target the underlying molecular changes involving oncogenes and tumor suppressor genes, genes that play critical roles in the conversion of normal cells into a cancerous state. EGFR belongs to a family of growth factor receptors which also includes HER-2/neu. It can\nheterodimerize with each of the members of this family.\n\nApplications: \nSuitable for use in Immunoprecipitation/Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWetern Blot: 1:2,000 to 1:3,000\nImmunoprecipitation:10ul/1ml of lysate (lysate ~1mg of total protein)\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: A431 cells, keratinocytes in normal epidermis or placenta.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
