Epithelial Sodium Channel, alpha (ENAC Alpha)

Tissue acidosis (decrease in pH below the physiological level) that occurs in ischemia, tissue damage or inflammation is accompanied by pain. At the molecular level, H+-gated cation channels are activated by low pH in nociceptive neurons. H+-gated cation channels, members of the NaC/DEG superfamily of Na channels that include amiloride-sensitive epithelial Na+ channel proteins (alpha, beta, gamma and delta-ENaC subunits), are expressed in epithelia of the vertebrate kidney, colon, lung, tongue and brain. The ENaC subunits may form heterotrimeric active sodium channel. ENaCs are involved in sodium and water readsorption, and salty taste transduction. The superfamily of DEG/NaC proteins are characterized by intracellular N and C-termini, two TM domains, and a large extracellular loop. \n\nENaC alpha subunits have been cloned from various tissues and from several species (human 669aa, rat 698aa, mouse 699aa; ~80% inter-species homology). It controls the reabsorption of sodium in the kidney, colon, lung and sweat glands. ENaC alpha has also been implicated in taste perception. Defects in ENaC alpha are one of the cause of pseudohypoaldosteronism type I, a rare salt wasting disease characterized by dehydration, hyponatacemia, hyperkalemia and acidosis. Inactivation of ENaC alpha in transgenic mice causes an early death due to defective lung liquid clearance.\n\nApplications: \nSuitable for use in ELISA and Western Blot. Note: Because of its low MW (< 3kD). The control peptide is not suitable for Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1-10ug/ml using ECL.\n\nELISA: 0.5-1ug/ml. Control peptide can be used to coat ELISA plates at 1ug/ml. See E3414-61F.\n\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes rat Epithelial Sodium Channel, alpha (ENAC alpha). The 20aa rat control peptide sequence homology is: 90% in mouse, 75% in human, 70% in rabbit, 65% in bovine. No significant homology is detected with ENAC beta, gamma or delta subunits.
