The human Epidermal Growth Factor Receptor (EGFR) family is a family of receptor tyrosine kinases consisting of 4 members: EGFR, HER2, HER3 and HER4. These receptors are expressed on a wide variety of tissue types and have been implicated in many human cancers. The ligand for HER3 and HER4 is called Neu Differentiation Factor (NDF) or Heregulin. In some instances, antibodies against these receptors prove effective in combination therapy regimens for treatment of the disease. Signaling through the EGFR family involves the combinatorial dimerization of the four known family members and is mediated by at least 8 different hormones, including EGF and the heregulins. Heregulin binding and tyrosine phosphorylation by HER3 or HER4 is enhanced in the presence of HER2. Structurally the members of the EGFR family are 1210-1342 amino acids in length with ~650 amino acids of the extracellular domain being heavily glycosylated. The extracellular domains show up to 70% sequence homology.\n\nApplications: \nSuitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nELISA: 1:10,000-1:50,000\nImmunoprecipitation: 5ul. Use with 25ul of Protein A-agarose beads and 1ml of lysate (lysate contains ~1mg of total protein). Rotate mixture overnight at 4C and then wash 3x with lysis buffer. Dissove the resulting bead complex in 20-30ul of 3xSDS-PAGE sample buffer and load ~15ul per lane on an 8% polyacrylamide gel.\nWestern Blot: 1:500-1:2000\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nHuman prostate cell lines (LNCap) or human breast cell line (MDA-453).\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
