c-erb B-3/HER-3 is a member of the EGF receptor tyrosine kinase family. It is a kinase defective receptor which signals by forming heterodimers with other erb family members. c-erb B-3 is over-expressed in a variety of tumors including breast, stomach, pancreas and colon, although the significance is unclear because over-expression of c-erb B-3 has no effect on tumorigenic growth of cells. Heregulin and EGF stimulate tyrosine phosphorylation of c-erb B-3 to different extents. This antibody can be used to assess the contribution of the other erb family members co-expressed with c-erb B-3 to ligand-dependent signaling because it inhibits the binding of heregulin (NDF) to c-erb B-3/HER-3.\n\nApplications: \nSuitable for use in Neutralization and Immunoprecipitation.\nOther applications not tested.\n\nRecommended Dilutions: \nNeutralization: 10ug/ml blocked heregulin mediated activation of c-erb B-3/HER-3 receptor in MCF-7 cells. Serum starved MCF-7 cells were stimulated with heregulin in the presence or absence of the neutralizing E3451-36. was immunoprecipitated with 5ug E3451-35. Immune complexes, split into duplicate samples, were resolved by electrophoresis and transferred to nitrocellulose. Duplicates were then treated with 1ug/ml P4078-05 or 1ug/ml E3451-35. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.\nImmunoprecipitation: Immunoprecipitates unoccupied c-erb B-3/HER-3 receptors.\nOptimal dilutions to be determined by the researcher.\n\nHybridoma:\nP3X63Ag8U1 (p3U1) cells with spleen cells from Balb/c mice. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
