ErbB4 (c-erbB4, HER4)

ErbB4 is a receptor tyrosine kinase that belongs to the human epidermal growth factor receptor (EGFR) family. ErbB4 is most predominantly expressed in several breast carcinoma cell lines, and in normal skeletal muscle, heart, pituitary, brain, and cerebellum. ErbB4 is activated by neuregulins (NRG), betacellulin (BTC), and heparin-binding EGF-like growth factor. Neuregulins regulate the expression of ligand- and voltage-gated channels in neurons and skeletal muscle by the activation of ErbB 1-4. The ErbB4 receptor is unique among these receptors because its C-terminal tail binds to a protein motif, known as the PDZ domain.\n\nApplications:\nSuitable for use in Immunoprecipitation, Biological Blockade of Heregulin Activity, Neutralization and Flow Cytometry. Not recommended for Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nImmunoprecipitation: 10ug immunoprecipitates erbB-4/HER-4 from 500ug of RIPA lysate prepared from hEGF stimulated A431 cells, as demostrated by subsequent immunoblot of the immunoprecipitate with P4078-05. The immune complex was subsequently resolved by electrophoresis and transferred to nitrocellulose and probed with P4078-05 at 1ug/ml. Proteins were visualized using a secondary antibody conjugated to HRP and a chemiluminescence detection system.\nBiological Blockade of Heregulin Activity: Reported to inhibit heregulin activity as measured by mitogenicity and radio-ligand binding assays. \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
