ErbB4 (c-erbB4, HER4)

erbB4/HER4 is the fourth member of the human epidermal growth factor receptor (EGFR) family that is frequently overexpressed in a variety of aggressive epithelial carcinomas. Its extracellular domain is most similar to the orphan receptor HER3, whereas its cytoplasmic kinase domain exhibits 79% and 77% identity with EGFR and HER2, respectively. HER4 is most predominantly expressed in several breast carcinoma cell lines, and in normal skeletal muscle, heart, pituitary, brain and cerebellum. Breast tumor cell lines MDA-MB-453, BT-474, T47-D and H3396 have been shown to have the highest levels of erbB-4 mRNA. Intermediate levels have been seen in MCF-7, MDA-MB-330 and MDA-MB-361 cell lines. erbB-4 expression has been shown to be low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468 and SKBR-3 cell lines. \n\nApplications: \nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1-4ug/ml detects erbB4/HER4 in RIPA lysates from MCF-7 and MDA-MB-453 cells. \nImmunoprecipitation: 4ug immunoprecipitates erbB4/HER4 from 500ug of MCF-7 RIPA lysate. \nImmunoprecipitation/Western Blot Analysis: MCF-7 cell lysate was immunoprecipitated with 4ug of E3451-41. The immune complex was subsequently resolved by electrophoresis and transferred to nitrocellulose. Proteins were visualized using a secondary antibody conjugated to HRP and a chemiluminescence detection system. \nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies:\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb\nI1904-46Q: IgG, H&L (HRP) Pab Gt xRb\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
