Erythropoietin Receptor (EPOr, EPO-R) (APC)

Erythropoietin R is a transmembrane protein expressed on the surface of megakaryocytes, erythroid progenitors, and endothelial cells. It binds Epo and transmits signals that stimulate the proliferation and maturation of bone marrow erythroid precursor cells into red blood cells.\n\nApplications:\nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 10ul labels upto 1x10e6 cells. Using TF-1 cells.\nOptimal dilutions to be determined by the researcher.\n\nExcitation: 620-650nm\n\nEmission: 660-670nm\n\nFlow Cytometry Protocol:\n1. Cells may be Fc-blocked with 1ug of human IgG/1x10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.\n2. After blocking, 10ul of E3455-23G was added to up to 1x10e6 cells and incubated for 30 minutes at RT.\n3. Unbound antibody was removed by washing the cells twice in F4568-10: Flow Cytometry Staining Buffer. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer.\n4. The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis (Excitation: 620-650nm, Emission: 660-670nm). As a control for this analysis, cells in a separate tube should be treated with APC-labeled mouse IgG1 antibody.\n\nStorage and Stability:\nMay be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C. Stable for 12 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap. reezing Labeled with Allophycocyanin (APC) conjugates will result in a substantial loss of activity. Further dilutions can be made in assay buffer. APC is light sensitive.
