Estrogen receptor-a is a ligand activated, transactivating protein. The wild type estrogen receptor-a coded by a 6.6kb mRNA, comprised of five functional domains derived from eight exons. The three domains of the protein which show the greatest degree of sequence homology are the A domain which, in concert with the B domain, contains the transactivation function (AF-1), the C domain which contains two zinc-fingers and is responsible for DNA binding and the E domain which is responsible for ligand binding as well as the ligand inducible transactivation function (AF-2). Amplification of the protein is a common mechanism in proliferative breast disease. Estrogen receptor-a IVS1-397T/C polymorphism is associated with risk of myocardial infarction and other cardiovascular disease in both men and women. Human estrogen receptor-a gene is mapped to chromosome 6q25.1. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Immunohistochemistry (paraffin): 1:50-1:100 Optimal dilutions to be determined by the researcher. Positive Control: MCF7 cells Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
