eIF4E is the smallest subunit of eIF4F, which is a supramolecular complex consisting of three subunits, eIF4E, eIF4A, and eIF4G, and catalysing the initiation of cap-dependent translation with a sequential function. eIF4E binds selectively to the cap structure and is a master switch that controls eukaryotic translation, because the binding of eIF4E to the cap structure is required for initiating the translation of mRNA and is regulated through the association/dissociation of eIF4E with endogenous 4E-binding proteins (4E-BPs). The protein also promotes cellular growth and transformation. The levels of eIF4E are used as prognostic indicators of clinical outcome in human cancers, including breast cancer and head and neck squamous cell carcinomas. Recent studies reveal that eIF4E also acts as a promoter of nucleocytoplasmic transport of specific transcripts Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Immunohistochemistry (paraffin): 1:50-1:100 Optimal dilutions to be determined by the researcher. Positive Control: NIH/3T3 cells Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
