F4/80 Antigen (APC)

F4/80 antigen is a member of the EGF-TM7 family of proteins which shares 68% overall amino acid identity with human EGF module-containing mucin-like hormone receptor 1 (EMR1).\nExpression of F4/80 is heterogenous and is reported to vary during macrophage maturation and activation. The F4/80 antigen is expressed on a wide range of mature tissue macrophages including Kupffer cells, Langerhans, microglia, macrophages located in the gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma and macrophages in the red pulp of the spleen. F4/80 expression has also been reported on a subpopulation of dendritic cells, but is absent from macrophages located in T cell areas of the spleen and lymph node. The ligands and biological functions of the F4/80 antigen have not yet been determined but recent studies suggest a role for F4/80 in the generation of efferent CD8 positive regulatory cells.\n\nApplications: \nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilutions:\nFlow Cytometry: Neat-1:10; 10ul labels 10e6 cells in 100ul.\nOptimal dilutions to be determined by the researcher.\n\nHybridoma:\nSpleen cells from immunized HOB2 rats were fused with cells of the mouse NS-1 mouse myeloma cell line.\n\nStorage and Stability:\nMay be stored at 4 degrees C before opening. DO NOT FREEZE! Stable for at least 12 months at 4 degrees C. Freezing allophycocyanin conjugates will result in a substantial loss of activity. Allophycocyanin conjugates are sensitive to light.

Recognizes the mouse F4/80 antigen, a 160kD glycoprotein expressed by most mouse macrophages. Binds to macrophages from different sites including the peritoneal cavity, lung, spleen and thymus, to blood monocytes and to macrophages derived from bone marrow precursors in culture. F4/80 antigen is not expressed by lymphocytes or polymorphonuclear cells.
