FADD (Fas-Associated Death Domain Protein, MORT1)

FADD was originally isolated as a protein that bound to the cytoplasmic domain of Fas in the yeast two-hybrid system. Sequence analysis revealed a region homologous to the death domain of Fas and TNFR-1. Subsequent biochemical studies have shown that FADD associates with Fas through interaction of the death domains. When over expressed in several cell lines, FADD induces apoptosis, which can be blocked by CrmA, an inhibitor of Caspase 1 (ICE). This evidence suggests that FADD plays a role in Fas-mediated apoptosis.\n\nApplications:\nSuitable for use in Immunoblot. Other applications have not been tested.\n\nRecommended Dilutions:\nImmunoblot: 0.5-2ug/ml. Detects FADD in RIPA lysated from Jurkat cells\nOptimal dilutions to be determined by researcher.\n\nPositive Control (included):\nJ9000: Jurkat Cell\n\nHybridoma:\nFusion of mouse myeloma P3U1 with Balb/c splenocytes.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.\n
