G Protein, alpha, (Goa)

GTP-binding proteins (G-proteins) play a critical role in signal transduction by coupling receptors to effectors. The intracellular consequences of G-protein activation include second messenger generation, protein phosphorylation, ion channel activation, gene induction, cell growth, and differentiation. Go belongs to the pertussis toxin sensitive G-proteins. It consists of two subtypes of a-subunits (ao1 and a2) derived from alternative splicing of a single gene. ao1 differs from ao2 within the C-terminal third of the deduced amino acid sequence. Some of the literature suggests that Transducin is the generic name for the heterotrimeric G protein or possibly Gt. Transducin is composed of several subunits. G proteins are further categorized into different classes, including Go and Gi, based on their aa sequence. Goa is a subunit of Go. Goa could be one subgroup of Transducin. However, there is some literature stating that Go, Gi, and Transducin are all separate groups of G proteins.\n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 1:3000. Detects a single protein band of 39-42kD in bovine and rat membranes.\nOptimal dilutions to be determined by researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes bovine G Protein, alpha, (Goa). Specificity confirmed by Western Blot versus bovine Gat, rat Goa, rat Gia1, Gia2 and Gia3; bovine Gsa; mouse GoaA and GoaB. The carboxy-terminal splice variants of Goa (GoaA and GoaB) expressed in bacterial lysates were compared for reactivity and found to be equal. As GoaA and GoaB have >95% homology to rat Goa1 and human Goa2. Species Crossreactivity: human; 70%, rat; 61%, mouse; 54%, guinea pig; 27%.
