GADD45 (Growth Arrest and DNA damage inducible) proteins are stress sensors and are implicated in stress response signaling that results in cell cycle arrest and DNA repair. The three GADD45 proteins (GADD45a, GADD45b, and GADD45g) are evolutionarily conserved with an approximate 50% homology. GADD45 is known to interact with a number of other nuclear proteins that include PCNA, p21/WAF/Cip, Cdc2, and the SAPK/MAPK pathway upstream of JNK and p38 by interacting with MKK4 and MKK7.
Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions:
Western Blot: 1:1000 detects GADD45a in 10ug of HEK293 lysates
Immunohistochemistry (Paraffin): 1ug/ml. Immunoreactivity is seen as membrane staining in human placental lining cells, pretreated with citrate buffer, pH 6.0.
Optimal dilutions to be determined by the researcher.
Positive Control:
HEK293, Lung tissue, Kidney tissue
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
