GAL4, DNA Binding Domain

GAL4 is a transcriptional activator that controls expression of the genes necessary for galactose catabolism. The DNA binding domain of GAL4, amino acids 1-147, binds to a specific 17-bp nucleotide sequence, but alone is insufficient to activate transcription. Residues 1-147 contain a zinc requiring DNA-binding motif and a region that mediates dimer formation.\n\nApplications:\nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: A 1:1000-5000 dilution detects GAL4 from a lysate prepared from insect cells infected with a baculovirus expressing GAL4. A 1:500 dilution of G1018 detected GAL4 (106kD) in a yeast cell extract. However, depending on the sample, additional bands may be observed. \nImmunoprecipitation: 1:1000 Immunoprecipitates GAL4 constructs from 35S-methionine labeled yeast and E. coli. Depending on the nature of the construct, phosphorylated and unphosphorylated forms of GAL4 may also be immunoprecipitated.\nOptimal dilutions to be determined by the researcher.\n\nRecommended Secondary Antibodies\nI1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb\nI1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb\nI1904-46Q: IgG, H&L (HRP) Pab Gt xRb\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
