The Fluorogenic GCN5 Assay Kit is a homogeneous, fluorogenic assay designed to screen for inhibitors of GCN5 or to measure histone acetyltransferase (HAT) activity using cell extracts or purified enzymes. It comes in a convenient 96-well format, with all the reagents necessary for 100 fluorescent HAT activity measurements. No time- consuming washing steps are required. In addition, the kit includes purified GCN5 (also known as KAT2A) for use as a positive control. The Fluorogenic GCN5 Assay Kit is based on the transfer of an acetyl group from acetyl CoA to a peptide substrate. After incubation with acetyl CoA and the substrate, the GCN5 enzyme generates acetylated H3 peptide and CoASH. The thiol groups of CoASH can be detected with fluorogenic reagent at excitation = 360nm and emission = 460nm.\n\nIntended Use:\nDesigned to screen for inhibitors of GCN5 or to measure histone acetyltransferase (HAT) activity using cell extracts or purified enzymes.\n\nTest Principle:\nThe Fluorogenic GCN5 Assay Kit is based on the transfer of an acetyl group from acetyl CoA to a peptide substrate. The thiol groups of CoASH can be detected with fluorogenic reagent at excitation = 360nm and emission = 460nm.\n\nKit Components:\nGCN5 recombinant human enzyme: 1x25g\nAcetyl CoA (1mM): 1x100ul\nHAT assay buffer (10X): 1x5ml\nH3 peptide: 1x500ul\nFluorescence Developer: 1x250ul\nDeveloper Solution: 1x10ml\nHAT Stop Solution: 1x5ml\nBlack, low binding NUNC black microtiter plate: 1 plate\n\nStorage and Stability:\nStore other components at 4 degrees C. Stable for 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
