Gelsolin is a calcium regulated actin binding protein. Gelsolin is a regulator of actin filament assembly and disassembly and consists of six homologous subdomains (S1-S6). When C-terminal tail (S6) is bound to calcium ion, it straightens and exposes actin binding sites located on S2s helices, leading to Gelsolin actin-serving activity (1). Gelsolin is inhibited by phosphatidylinositol (4,5)-bisphosphate (PIP2), where PIP2 will bind to S1, S2, and S3 sites. Gelsolin is involved in podosome formation and has been linked to inhibition of apoptosis by preventing release of cytochrome C (2). Applications: Suitable for use in Flow Cytometry, Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:100- 1:500 Western Blot: 1:50,000-1:200,000 Immunohistochemistry: 1:250-1:500 Immunocytochemistry: 1:100-1:250 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
