PKCq is a member of the novel, Ca(2+)-independent PKC subfamily, which plays an important and non-redundant role in several aspects of T cell biology. Mutation of PKCq gene leads to impaired receptor-induced stimulation of the transcription factors AP-1, NF-kappaB and NFAT, which results in defective T cell activation, and to aberrant expression of apoptosis-related proteins, resulting in poor T cell survival. It is the major isoform of PKC that is involved in NF-kB activation induced by CD3-CD28 costimulation. PKCq is expressed mostly in skeletal muscle, megakaryoblastic cells and platelets. Human PKCq gene is localized on human chromosome 10p15. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:500-1:1000 Immunohistochemistry (paraffin): 1:50-1:100 Optimal dilutions to be determined by the researcher. Positive Control: HT-29 and K562 cells Storage and Stability: May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
