The glucocorticoid receptor (GR) belongs to a superfamily of transcription factors that includes receptors for steroid and thyroid hormones, retinoic acid and vitamin D3. GR is normally localized in the cytoplasm, in a non-active state, in a complex with Hsp90 and other factors. On hormone binding, GR changes conformation, is released from the complex, and migrates to the nucleus. Once in the nucleus, the receptor can induce or repress transcription by binding to specific DNA sequences on target genes. The GR has a ubiquitous expression pattern in mammals and is involved in regulation of a number of biological processes through regulation of various target genes. In the mammary gland, it is essential for the differentiation process that leads to lactation.
