Glutamate is a major excitatory neurotransmitter. One enzyme central to the metabolism of glutamate is glutamate dehydrogenase GDH1, that catalyzes the reversible deamination of L-glutamate to 2-oxoglutarate using NAD+ or NADP+. Mammalian GDH is composed of six identical subunits, and the regulation of GDH is very complex. It has been a major goal to identify the substrate and regulatory binding sites of GDH. It is only in recent years that the three-dimensional structure of GDH from microorganisms is available. Very recently, crystallization of bovine liver GDH was reported for the first time from the mammalian sources. However, remarkably little is known about the detailed structure of mammalian GDH, especially the brain enzymes. Bovine glutamate dehydrogenase exists as a homohexamer located within the mitochondrial matrix.\n\nApplications: \nSuitable for ELISA, Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilutions: \nELISA: 1:4000-1:16,000. Sandwich ELISA coat plates with 1ug glutamate dehydrogenase and use peroxidase-conjugated goat anti-rabbit IgG.\nWestern Blot: 1:1000-1:3000 Expect a band ~56kD corresponding to glutamate dehydrogenase monomer\nsubunit in the appropriate cell or tissue extract.\nImmunoprecipitation: 1:100\nOptimal dilutions to be determined by the researcher. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes bovine Glutamate Dehydrogenase (GDH) at ~56kD. Crossreactivity is expected for mitochondrial and brain isoforms of GDH (GDH1 and GDH2) from both bovine and human sources based on sequence homology. Assay by IEP resulted in a single precipitin arc against purified and partially purified bovine liver glutamate dehydrogenase. Species sequence homology: Most primate species (including green monkey, white gibbon, chimpanzee orangutan and gorilla), chicken, mouse, rat, canine and other mammals as well as Xenopus tropicalis, zebrafish, rainbow trout and Atlantic salmon is expected. Crossreactivity with GDH from other tissues and species may occur.
