Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was considered a classical glycolytic protein involved exclusively in cytosolic energy production. However, recent evidence suggests that it is a multifunctional protein displaying diverse activities distinct from its conventional metabolic role. These new roles for GAPDH may be dependent on its subcellular localization, oligomeric state or on the proliferative state of the cell. Results suggest that human cells contain significant intracellular levels of enzymatically inactive GAPDH which is age-independent. The possibility is considered that the functional diversity of GAPDH may be mediated either by posttranslational alteration or by subcellular protein:protein and/or protein:nucleic acid interactions (1). GAPDH mRNA levels, protein, and enzymatic activity have been shown to increase increase in 3T3 cells after exposure to physiological concentrations of insulin (2). Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000-2000 (mouse samples: 1:500-1000) Immunohistochemistry: 1:100-250 Immunocytochemistry: 1:50-100 Immunoprecipitation: 1:40 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
