Glycogen synthase 3 alpha and beta (GSK-3alpha and GSK-3beta) are two highly related proteins of 51 and 47kD that function as broad specificity serine/threonine kinases (1). Although they are the products of distinct genes, GSK 3alpha and GSK3beta are 93% identical over the catalytic domain, with GSK3 alpha containing an additional 40 residues (2). The two isoforms are ubiquitously distributed, with the highest levels of expression occurring in the brain (2). GSK-3alpha is identical to multifunctional ATP-citrate lyase kinase and GSK3beta is identical to tau protein kinase 1 (TPK1) (1,3). GSK3beta is believed to be involved in the hyperphosphorylation of tau protein in the Alzheimer diseased brain, and in the regulation of binding of beta catenin to APC (4,5). Applications: Suitable for use in Western Blot, Immunohistochemistry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
