Glycogen Synthase Kinase 3 alpha, Fusion Protein, Recombinant (GSK3a)

GSK-3 is one of the Akt substrates. It can be phosphorylated by Akt at Ser21 of GSK-3a or Ser9 of GSK-3b. The phosphorylation of GSK negatively regulates its kinase activity. The recombinant GSK-3a fusion protein serves as a useful substrate for assaying Akt kinase activity.\n\nGSK-3a was expressed in E. coli. It is expressed as a recombinant protein fusion of GSK-3a residues surrounding GSK-3a (Ser21) and glutathione S-transferase (GST). The protein was then puri?ed by af?nity chromatography.\n\nActivity: \nGSK-3a fusion protein at a concentration of 2ug/20ul reaction was phosphorylated in an in vitro kinase assay with 200 mM ATP. After 30 minutes incubation at 30 degrees C, phosphorylation was detected by Western blot with phospho-speci?c GSK-3a antibodies.\n\nApplications: \nSuitable for use in ELISA, Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 2ug/20ul\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nAliquot to avoid repeated freezing and thawing. Store at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
