Glycogen synthase kinase 3 (GSK3) is a serine/threonine kinase that is involved in the regulation of many signaling pathways. To date, 2 isoforms have been identified: GSK3a and GSK3b. Specifically, GSK3b has been shown to play a key inhibitory role in both the insulin and Wnt signaling pathways (Papkoff and Aikawa 1998). It has been suggested that Ser9 phosphorylation underlies the inhibition of GSK3b by insulin (Sutherland et al., 1993). Positive Control: Rat cortex lysate Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional info: The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with human, mouse, chicken, bovine, canine, non-human primate, Zebrafish and Xenopus based on the fact that these species have 100% homology with the amino acid sequence used as antigen. Specific for the ~46k GSK3b protein phosphorylated at Ser9. Also weakly labels the ~51k GSK3a band due to the high degree of homology between the 2 subunits. Immunolabeling is blocked by the phosphopeptide used as antigen but not by the corresponding dephosphopeptide.
