Glycoprotein 340 (gp340)

Glycoprotein (gp) 340 is member of the scavenger receptor superfamily and has been identified as a putative SP-D receptor. Gp340 has been shown to bind SP-D in a calcium-dependent manner through a protein-protein interaction (1). Like SP-D, gp340 is found in the lungs and immunohistochemistry analysis shows high expression in alveolar macrophages (1). An identical protein, salivary agglutinin, is present in saliva (2-4) and both gp340 and salivary agglutinin are encoded by the gene DMBT1 (5-7).\n\nApplications: \nSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nELISA: A strong reaction is seen when gp340 is directly coated onto the microtiter well.\nWestern Blot: 1:200-1:10,000 (1-4, 6). Strongly reacts with gp340 in both reduced and non-reduced forms in Western Blot after SDS-PAGE.\nImmunohistochemistry: see references 1,5,6\nOptimal dilutions to be determined by the researcher.\n\nCulture Medium: \nRMPI 1640 with 10% fetal calf serum.\n\nHybridoma: \nX63-Ag8.653 myeloma cells with spleen cells from Balb/c mice. \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
