Goserelin, Human

Goserelin has actions similar to those of naturally occurring LHRH. LHRH released from the hypothalamus stimulates receptors on the pituitary gland, which then releases the gonadotropins luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Goserelin and other LHRH agonists provide a medical castration that deprives hormonally-dependent tumors of testosterone or estrogen. Normally, LHRH is released in a pulsatile manner to maintain levels of gonadotropins. Goserelin, in contrast, is continuously administered, which leads to downregulation of the LHRH receptor on the pituitary gland and decreased production of FSH and LH. During short-term or intermittent treatment, goserelin has the same stimulatory action as LHRH. This stimulation produces a surge of FSH and LH, which can cause a flare reaction in patients with prostate or breast carcinoma.\n\nSequence:\nGlu-His-Trp-Ser-Tyr-D-Ser(tBu)-Leu-Arg-Pro-AzGly-NH2\n\nEndotoxin:\nLess than 0.1ng/ug (IEU/ug) of Goserelin.\n\nReconstitution:\nReconstitute the lyophilized Goserelin in sterile 18MOmega-cm H2O not less than 100microg/ml, which can then be further diluted to other aqueous solutions.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile dH2O and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
