HAMA (Human anti-Mouse Antibody) BioAssay(TM) ELISA Kit

Principle:\nThis assay applies an enzyme-linked immunosorbent assay (ELISA) technique. A purified mouse IgG has been pre-coated onto the wells of microplate. Standard (rabbit anti-mouse IgG) and specimens are added into the appropriate wells simultaneously with a HRP-labelled mouse IgG, and incubated. The Human Anti-Mouse Antibody (HAMA), if present, will bind with the mouse IgG which coated on the wells and HRP- labelled mouse IgG as well, forming a "Sandwich". Following a wash to remove any unbound substance, a substrate solution is added. The developed colour is directly proportional to the anti-mouse antibody concentration in the specimen and Standard samples.\n\nThis ELISA Kit includes a Calibration Diluent for preparing a 2-fold dilution of Standard series. It allows the operator to produce a standard curve with Optical Density (O.D) versus Human Anti-Mouse Antibody concentration (ng/mL). The concentration of Human Anti-Mouse Antibody in the sample is then determined by comparing the O.D. of the sample to the standard curve.\n\nKit Components:\nH1776A: HAMA Microtiter Plate, Pre-coated with mouse IgG. 1x96 wells\nH1776B: HRP Conjugate, Mouse IgG labeled with Horseradish Peroxidase (HRP). 1x12ml\n*H1776D: HAMA Standard, Rabbit anti-Mouse IgG antibody, supplied as a lyophilized powder from a buffered protein base with preservative. 2x800ng\nH1776E: Calibrator Diluent, Animal serum with buffer and preservative. 1x25ml\nH1776F: Wash Buffer (20X), 20-fold concentrated solution of buffered surfactant. 1x60ml\nH1776G: Substrate A, Buffered solution with H2O2. 1x10ml\nH1776H: Substrate B, Buffered solution with TMB. 1x10ml\nH1776J: Stop Solution, 2N Sulphuric Acid (H2SO4). 1x15ml\n\nStorage and Stability:\nStore *H1776D powder at 4 degrees C liquid at -20 degrees C. Store other components at 4 degrees C. Stable for 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
