HAPLN4 (Hyaluronan and Proteoglycan Link Protein 4, Brain Link Protein 2)

Hyaluronan and Proteoglycan Link Protein 4, (HAPLN4), also known as brain link protein 2, is one of four secreted HAPLN family proteoglycans that bind hyaluronan. HAPLN4 contains one Ig-type C2-set domain and two link domains. It is predominantly expressed in brain and participates in the development of the perineuronal matrix. Human and mouse HAPLN4 share 91% aa sequence identity.\n\nHybridoma: \nProduced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, NS0-derived, recombinant human HAPLN4 (rhHAPLN4; aa 1-402: Accession #NP_075378).\n\nApplications: \nWestern Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect human HAPLN4. Using a colorimetric detection system, the detection limit for rhHAPLN4 is approximately 1 ng/lane and 25 ng/lane under non-reducing and reducing conditions, respectively. Chemiluminescent detection will increase sensitivity by 5 to 50 fold.\nDirect ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human HAPLN4. The detection limit for rhHAPLN4 is approximately 5 ng/well. \nOptimal dilutions should be determined by each laboratory for each application.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
