HDAC1 (Histone Deacetylase 1)

Acetylation and deacetylation of lysine residues of core histones by histone acetyltransferases and HDACs respectively have been shown to be necessary for some crucial nuclear events such as gene transcription, DNA replication and repair. The current model is that acetylation affects the stability of nucleosomal structure and modulates the affinity of trans-acting factors. Recent biochemical and genetic research indicated that hyperacetylation of core histones is a characteristic of gene activation, while histone deacetylation is involved in transcriptional silencing. Mouse HDAC 1 is a homolog of the yeast transcription co-regulator RPD3 and has been indicated to be a transcription co-repressor. \n\nApplications:\nSuitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 0.5ug/ml lot detects HDAC 1in RIPA lysates from 3T3 cells. 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with H1827-50 (0.5ug/ml). Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system.\nImmunocytochemistry: This antibody has been reported to immunostain HDAC 1 in 1% paraformaldehyde fixed, 0.1% Triton X-100 permeabilized 3T3 cells. \nImmunoprecipitation: 5ug immunoprecipitates HDAC 1 from 500ug of 3T3 RIPA lysate. This antibody has been reported to Immunoprecipitate active HDAC.\nOptimal dilutions to be determined by the researcher.\n\nPositive Antigen Control: \nF4209: Fibroblast Cell, 3T3/A31. Add 2.5ul 2-mercaptoethanol/100ul of lysate. Boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
