HtrA3 (High Temperature Requirement Factor A3, HtrA Serine Peptidase 3, Calpain 5, CAPN5, EC 3.4.21., Pregnancy-related Serine Protease, Probable Serine Protease HTRA3, PRSP, Tasp)

HTRA3 Serine protease that cleaves beta-casein/CSN2 as well as several extracellular matrix (ECM) proteoglycans such as decorin/DCN, biglycan/BGN and fibronectin/FN1. Inhibits signaling mediated by TGF-beta family proteins possibly indirectly by degradation of these ECM proteoglycans. May act as a tumor suppressor. Negatively regulates, in vitro, trophoblast invasion during placental development and may be involved in the development of the placenta in vivo. May also have a role in ovarian development, granulosa cell differentiation and luteinization. Belongs to the peptidase S1B family. 2 isoforms of the human protein are produced by alternative splicing. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
