Hyaluronidases (HAases)1 are a family of enzymes that are crucial for the spread of bacterial infections, toxins present in various venoms, and possibly, cancer progression.1-6 In humans, six HAase genes have been identified. With the possible exception of HYAL4 and HYALP1, all other HAases are thought to degrade hyaluronic acid (HA).7 HA is a high molecular mass nonsulfated glycosaminoglycan made up of repeating disaccharide units, D-glucuronic acid, and N-acetyl-D-glucosamine. HA is present in body fluids, tissues, and the extracellular matrix of vertebrate tissues.8-10 It keeps tissues hydrated, and maintains osmotic balance, and cartilage integrity.8,9 HA also actively regulates cell adhesion, migration, and proliferation by interacting with specific cell surface receptors such as CD44 and RHAMM.11 The concentrations of HA are elevated in several inflammatory diseases, and various carcinomas such as bladder, prostate, breast, lung, and colon.8,12-19 HYAL3 transcripts have been detected in kidney, brain and liver tissues.20 HYAL3 has a classical TIM barrel topology, where the catalytic site corresponds to Asp127 and Glu129, respectively. Several mRNA splice variants of HYAL3 expressed in bladder and prostate cancer cells and tissues have been identified and characterized in terms of their protein product and HAase activity.21 A 30-aa region that is crucial for the HAase activity of HYAL3 protein was also identified.21\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation. Other applications not tested.\nRecommended Dilution:\nWestern Blotting: 1-3ug/ml\nImmunoprecipitation: 5ug/reaction\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n\n

Reacts with the mouse Hyal3 protein. On Western blots of cells overexpressing Hyal3, the target band is identified at ~50kD. Reactivity has been confirmed with mouse Hyal3-transfected BHK cell lysates.
