ICAD (DNA Fragmentation Factor Subunit alpha, DNA Fragmentation Factor 45kD Subunit, DFF-45, Inhibitor of CAD, ICAD, DFFA, DFF1, DFF45, H13)

Apoptosis is related to many diseases and is induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These cell death signals cause the degradation of chromosomal DNA by activated DNase. A human DNA fragmentation factor (DFF) was identified recently which was cleaved by caspase-3 during apoptosis. The mouse homologue of this human DFF was identified as a DNase inhibitor designated ICAD. Upon cleavage of DFF/ICAD, a caspase activated deoxyribonuclease (CAD) is released, activated and eventually causes the degradation of DNA in the nuclei. Therefore, the cleavage of CAD inhibitor molecule DFF/ICAD, which causes DNase activation and DNA degradation, is identified as a hallmark of apoptotic cell death.\n\nApplications:\nSuitable for use in Western Blot and Immunocytochemistry. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:500\nImmunocytochemistry: 1:500\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nUntreated and staurosporin-treated HeLa cell lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
