IL6R (Interleukin-6 Receptor Subunit alpha, IL-6 Receptor Subunit alpha, IL-6R Subunit alpha, IL-6R-alpha, IL-6RA, IL-6R 1, Membrane Glycoprotein 80, gp80, CD126) (APC)

IL6R is a transmembrane protein that associates with the signal-transducing subunit gp130 to form a high-affinity receptor complex for IL-6. It is expressed on T cells, activated B cells, and peripheral monocytes. A soluble form of IL6R can be released by proteolytic cleavage.\n\nApplications:\nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilutions:\nFlow Cytometry: 10ul labels 1-2.5x10e5 cells. Validated using CD3+ mouse splenocytes.\nOptimal dilutions to be determined by the researcher.\n\nFlow Cytometry Protocol:\n1. Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at RT. Do not wash excess blocking IgG from this reaction.\n2. After blocking, add 10ul of I8428-39M2 to 1-2.5x10e5 cells and incubate for 30 minutes at RT.\n3. Remove unbound antibody by washing the cells twice in Flow Cytometry Staining Buffer (F4568-10). Note that whole blood requires a RBC lysis step at this point using Flow Cytometry mouse lyse buffer.\n4. Resuspend the cells in Flow Cytometry Staining Buffer for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with APC-labeled rat IgG2a antibody.\nNote: The procedure may need to be modified, depending on the cell type and final utilization.\n\nStorage and Stability:\nMay be stored at 4 degrees C before opening. DO NOT FREEZE! Stable at 4 degrees C as an undiluted liquid. Stable for at 12 months at 4 degrees C. Freezing allophycocyanin conjugates will result in a substantial loss of activity. Allophycocyanin conjugates are sensitive to light.
