Immunoassays using enzyme labeled antibodies are highly specific for the analysis of a particular protein. Use of an enzyme labeled antibody together with a highly sensitive TMB substrate provides an excellent method for the detection and characterization of sample proteins bound to solid surface by ELISA techniques. Following attachment of protein to the solid surface (ELISA plate), a primary antibody is used to selectively bind the protein of interest. Alternatively, a known protein is bound to the surface for the screening of specific monoclonal/polyclonal antibodies in serum or other samples. An enzyme labeled second antibody directed against the primary antibody is then applied. The TMB substrate reacts with bound HRP to produce blue color. Upon addition of stop solution, the color is converted to yellow color (measure at 450nm). The yellow color is more intense and thus more sensitive than the blue color. Intensity of color, up to a certain range, is directly proportional to the amount of bound primary antibody.\n\nThis ELISA kit is a qualitative, direct antigen-based assay designed to detect the presence of hamster antibodies in serum and to determine antibody titer using antigen-coated plates. It is not suitable for the quantitation of hamster IgG. The HRP-conjugated anti-hamster IgG provided in this kit is made against highly pure, whole antigen molecule specific to both H and L chains and will detect all IgG's (IgG + IgA + IgM). The reagents contained in this kit are sufficient for 1000 to 2000 samples or 10-20 plates.\n\nKit Components:\nI7502-10A: IgG (HRP) anti-hamster 1x150ul. \nI7502-10B: Coating Buffer, 10X, 1x25 ml. \nI7502-10C: Blocking Buffer, 10X, 1x25ml. \nI7502-10D: Sample/Conjugate Diluent, 10X, 1x25ml. \nI7502-10E: Wash Buffer, 100X, 1x25ml.\nI7502-10F: TMB Substrate, 1x100ml.\nI7502-10G: Stop Solution, 10X, 1x10ml. \n\nStorage and Stability:\nStore all kit components at 4 degrees C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
