Isocitrate Dehydrogenase, Recombinant (ICDH)

Isocitrate Dehydrogenase is an enzyme of the oxidoreductase class that catalyzes the conversion of isocitrate and NAD+ to yield 2-ketoglutarate, carbon dioxide, and NADH. It occurs in cell mitochondria. The enzyme requires Mg2+, Mn2+; it is activated by ADP, citrate, and Ca2+, and inhibited by NADH, NADPH, and ATP. The reaction is the key rate-limiting step of the citric acid (tricarboxylic) cycle.\n\nDimers and Aggregates: \n(same/less than) 1% as determined by silver-stained SDS-PAGE gel analysis. Endotoxin: (same/less than) 0.1ng/ug (IEU/ug) of riCDH.\n\nUnit Definition: \nOne unit is defined as 1 mol of NAD+ production per minute under the assay conditions (25 degrees C, pH 7.5).\n\nSpecific Activity:\n(same/more than) 40U/mg protein (at 25 degrees C and pH 8.5)\n\nContaminant: \n(as rICDH(NAD+) activity = 100%) \nIsocitrate dehydrogenase (NADP+) \n\nApplications:\nSuitable for use as a calibrator or control serum. \n\nReconstitution:\nReconstitute with sterile Tris-HCl, pH8.5 to a concentration of 0.1-1mg/ml. \n\nStorage and Stability:\nAliquot to avoid repeated freezing and thawing and store at -70 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 6 months.
