Human DcR3 (decoy receptor 3), also known as TR6, M6, and TNFRSF6, is a soluble member of the TNFR superfamily. It acts as a decoy receptor by competitively binding FasL, LIGHT, and TL1A away from their individual receptors, thereby neutralizing their effects. Soluble DcR3 also acts as an effector molecule by directly causing immunomodulation. For example, it suppresses Th1 formation and skews CD4+ T cells towards a Th2 phenotype.DcR3 is a 34 kD protein consisting of 300 amino acids. It possesses four TNFR domains containing four cysteine-rich repeats. The crystal structure of free DcR3 reveals a dimer via weak hydrophobic interactions, whereas the DcR3-TL1A complex shows a strong 3:3 stoichiometric relationship between receptor and ligand. It shares ~30% amino acid sequence homology with OPG and TNFR2. There are no homologs in mice or rats.Soluble DcR3 has been implicated in various inflammatory, auto-immune, and neoplastic conditions. It may be useful as a cancer biomarker, since serum levels of soluble DcR3 are elevated in certain cancer patients compared to normal. Its levels also seem to correlate with disease progression and prognosis.
