Mannan Binding Protein (MBP, Mannose Binding Protein, Mannan Binding Lectin, MBL)

Mannan-binding lectin (MBL) belongs to the C-type family of collectins. On specific binding to microbial surface carbohydrates, MBL activates the complement system by means of its own lectin pathway, depending on the MBL-associated serine proteases (MASPs). Investigations have shown that up to 12% of normal blood donors (2) have low (<50ng/ml) MBL plasma concentrations.\n\nApplications: \nSuitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilution:\nELISA: 1:10,000\nWestern Blot: 1:500\nImmunohistochemistry: Frozen or paraffin from liver and brain.\nOptimal dilutions to be determined by the researcher.\n\nHybridoma:\nX63-Ag8.653 myeloma cells with spleen cells from Balb/c mice.\n\nCulture Medium:\nRPMI 1640 with 10% fetal calf serum\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes human MBL from serum or plasma. Epitope specificity differs from those of clones 6F141 and 6F142. In Western Blot reacts with human MBL both in its polymeric conformation and as a single subunit of molecular mass 26kD. Used as detection antibody in sandwich ELISA with a mannan or clone 6F140 coat, clone 6F140 is selective for normally oligomerized MBL.
