p38 MAP kinase (MAPK) is also called RK (3) or CSBP (2). It is the mammalian homologue of the yeast HOG kinase. It participates in a signaling cascade controlling cellular responses to cytokines and stress (1-4). Four isoforms of p38 MAP (kinase, alpha, beta, gamma and delta) have been identified. Like the SAPK/JNK pathway, p38 MAP kinase is activated by a variety of cellular stresses including osmotic shock, inflammatory cytokines, lipopolysaccharides (LPS), UV light and growth factors (1-5). MKK3 and SEK activate p38 MAP kinase by phosphorylation at threonine 180 and tyrosine 182. Activated p38 MAP kinase has been shown to phosphorylate and activate MAPKAP kinase 2 (3) and to phosphorylate the transcription factors ATF-2 (5), Max (6) and MEF2 (5-8).\n\nApplications: \nSuitable for use in ELISA, Western Blot and Flow Cytometry. Other applications not tested.\n\nRecommended Dilution: \nWestern Blot: 1:2000. Incubate membrane with diluted M2352-01 in 1X TBS, 5% nonfat dry milk, 0.1% Tween-20 at 4 degrees C with gentle shaking, overnight.\nFlow Cytometry: 1:2000\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability: \nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
