Recognises human Neurite Outgrowth Inhibitor (NogoA), also known as Reticulon-4 (RTN4), a member of a family of integral membrane proteins termed reticulons. Research indicates that reticulons are involved in various neurodegenerative diseases. Reticulon proteins have been demonstrated to regulate many cellular processes and interact with multiple proteins and receptors such as BACE. NogoA was initially identified as a myelin-associated neurite outgrowth inhibitor. It is highly expressed in oligodendrocytes in the white matter of the CNS. Experimentation by blocking its activity with antibodies or other factors results in improved axon regrowth and functional recovery in experimental CNS lesion models. NogoA has also been suggested to play a role in neurodegenerative diseases such as Amyotrophic lateral sclerosis,and multiple sclerosis. Despite its predicted molecular weight, NogoA typically migrates at 180kD in an SDS-PAGE. Five isoforms of Nogo have currently been identified. \n\nApplications:\nSuitable for use in Western Blot and Immunohistochemistry. Other applications have not been tested. \n\nRecommended Dilutions: \nWestern Blot: 0.5-1ug/ml detects a band of ~180kD in Mouse Brain tissue lysate.\nImmunohistochemistry (paraffin): 2.5ug/ml \nOptimal dilutions to be determined by the researcher. \n\nImmunohistochemistry Positive Control:\nMouse brain\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
