Neurogranin is a calmodulin-binding protein expressed exclusively in the brain, particularly in dendritic spines, and participating in the protein kinase C signaling pathway. Neurogranin is the main postsynaptic protein regulating the availability of calmodulin, binding to it in the absence of calcium. Phosphorylation by protein kinase C lowers its binding ability. NRGN gene expression is controlled by thyroid hormones. Human neurogranin consists of 78 amino acids.\nOne study tells of potential link of neurogranin gene to the heightened risk of schizophrenia in males, another study gives evidence of lowered neurogranin immunoreactivity in the brains of people suffering from schizophrenia.Prior to its identification in the bovine and rat brain in 1991, neurogranin was known as a putative protein kinase C-phosphorylated protein named p17.\n\nIntended use:\nNeurogranin (NRGN) BioAssay(TM) ELISA Kit (Human) is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of human NRGN in tissue homogenates, cell lysates and other biological fluids.\n\nTest Principle: \nThis assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific for NRGN has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled NRGNand unlabeled NRGN(Standards or samples) with the pre-coated antibody specific for NRGN. After incubation, the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of NRGN in the sample. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.\n\nAssay Procedure Summary:\n1. Prepare all reagents, samples and standards. \n2. Add 50ul standard or sample to each well. And then add 50ul prepared Detection Reagent A immediately. Shake and mix. Incubate 1 hour at 37 degrees C. \n3. Aspirate and wash 3 times. \n4. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37 degrees C. \n5. Aspirate and wash 5 times. \n6. Add 90ul Substrate Solution. Incubate 15-25 minutes at 37 degrees C. \n7. Add 50ul Stop Solution. Read at 450nm immediately.\n
