Neutrophils (Ly-6G NEUTROPHIL MARKER, Gr1, Gr-1, Ly-6G, Ly-6G.1, Lymphocyte antigen 6G precursor, Lymphocyte antigen Ly-6G.1 precursor) (Biotin)

Ly6G is together with Ly6c a component of the myeloid differentiation antigen Gr-1. Ly6G is a GPI- anchored protein and a good marker of peripheral neutrophils. Although predominantly present on neutrophils, it is also expressed on a subset of eosinophils, differentiating pre-monocytes and plasmacytoid dendritic cells. Ly6C is a monocyte/macrophage and endothelial cell differentiation antigen regulated by interferon gamma, and may play a role in the development and maturation of lymphocytes. It is expressed on bone marrow cells, monocytes/macrophages, neutrophils, endothelial cells and T cell subsets. N2254-01 has been successfully used to stain polymorphonuclear (PMN) cells and monocytes for fluorescent activated cell sorting and in frozen and paraffin sections. Treatment with antibodies in vivo leads to neutropenia and has an inhibitory effect on local immune responses. Furthermore, it has been shown to be useful for depletion of neutrophils in mice. It depletes neutrophils as soon as 6 hours after injection and up to 6 days.\n\nApplications:\nSuitable for use in ELISA, Flow Cytometry, Functional Studies and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nFlow Cytometry: 1:50 (10ug/ml) labels 5x10e5 cells (4)\nImmunohistochemistry (Frozen): 1:50 acetone fixation is recommended. (7)\nImmunohistochemistry (Paraffin): 1:50 blocking with 20% normal rabbit serum. (5)\nFunctional Studies (remove sodium azide): Neutrophil depletion. Mice were tested with N2254-01 given intraperitoneally at a dose of 1mg, 6 hours before injection. (6)\nOptimal dilutions to be determined by the researcher.\n\nRecommended Positive Control: Mouse neutrophils\n\nRecommended Negative Control: Mouse Thymocytes\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
