Nibrin (Nijmegen Breakage Syndrome, NBS1)

Applications: \nSuitable for use in Western Blotting, Immunoprecipitation\n\nImmunoblot Analysis: 2ug/ml detects Nibrin/Nbs1 in Jurkat, HeLa nuclear extract, HeLa whole cell, and A431+/-EGF lysates. \n\nIncluded Positive Antigen Control: Jurkat lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. \n\nImmunoprecipitation: 4ug immunoprecipitats Nibrin/Nbs1 from 500ug of Jurkat RIPA lysate. Jurkat cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti- Nibrin/Nbs1 (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Nibrin/Nbs1 (95kD). \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
