Nkx 2.8 Mid (Nk2 Transcription Factor Related Locus 8, NKX2H, NKX2.8, Nkx2-9)

Nkx2.8 is part of a family of vertebrate developmental regulators that are homologues of the Drosophila homeodomain transcription factor, NK2. Nkx2.8 transcripts were readily shown in human fetal liver, and significant levels of Nkx2.8 protein were present in human, rat, and mouse hepatocellular carcinoma cell lines that express a-fetoprotein (AFP).1 Moreover, mechanistic studies in human hepatocellular carcinoma line HuH7 established that Nkx2.8 functions as a significant transcriptional activator that regulates (AFP) in vivo.2 The Nkx2.8 knockout mice exhibited late gestational changes in the central nervous system (CNS), suggesting its role in specifying neuronal cell fates along the dorsoventral axis within the CNS.3 Moreover, the studies demonstrated that Nkx2.8 was most strongly expressed in cells that function as tracheobronchial stem cells, with Nkx2.8 acting to limit this population. Targeted inactivation of the gene caused expansion of this cell population and widespread dysplasia.4 All these data suggest that Nkx2.8 plays important roles during CNS, lung and pharyngeal development.3,4\n\nApplications: \nSuitable for use in ELISA, Western Blotting, Immunoprecipitation. Identifies the target band at ~35kD.\nImmunoprecipitation: 5ug/reaction\nOptimal dilutions to be determined by the researcher.\n\nWB Positive Control:\nPurified protein with GST Tag (M.W. 61.38kD)\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
