Applications: \nSuitable for use in ELISA, Immunohistochemistry, Immunocytochemistry and Western Blot. Other applications not tested.\n\nRecommended Dilutions:\nImmunohistochemistry: 1:200-1:1000\nImmunocytochemistry: 1:200-1:1000\nWestern Blot (ECL): 1:200-1:1000\nOptimal dilutions to be determined by the researcher.\n\nImmunohistochemistry Protocol:\n1. Dissect tissues and freeze on dry ice \n2. Cut on a cryostat -10um slices \n3. Dry slides at RT for 30-90 minutes\n4. Fix with cold Acetone/Methanol (50/50) 2 minutes - then either process further or store at -20 degrees C . Alternative is fixation with 4% PFA and treatment with a trypsin solution. (0.05% trypsin, incubation 10 minutes at 37 degrees C, followed by 3 washes with PBS\n5. Let air dry \n6. PBS 5 minutes \n7. Incubate in 50mM ammonium chloride 30 minutes \n8. PBS 5 minutes \n9. Blocking serum 30-45 minutes \n10. N2851 90 minutes - dilution 1:200 - 1:1000 \n11. Wash 3 times for 5 minutes \n12. Secondary fluorescent conjug. antibody 30 minutes \n13. Wash 3 times for 5 minutes and mount\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
