NKX2.2 (Homeobox protein NKX)

The NKX families of homeobox genes are known to act as intermediaries in the neural response to sonic hedgehog signaling during central nervous system development. NKX2.2 is a member of this family of transcription factors and is necessary for neuroendocrine differentiation in the central nervous system and pancreas (Sander M, 2000). This protein was recently identified as a target of EWS-FLI-1, the fusion protein specific to Ewing sarcoma, and was shown to be differentially upregulated in Ewing sarcoma on the basis of array-based gene expression analysis. It also acts as a valuable marker for Ewing sarcoma, with a sensitivity of 93% and a specificity of 89%, and aids in the differential diagnosis of small round cell tumors (Yoshida A, 2012). NKX2.2 preferentially binds the promoter of the acell-specific factor Arx in bcells when the promoter is in a hypermethylated state. Disruption of the interaction between NKX2.2 and Grgs in mice leads to ectopic expression of Arx in bcells and is sufficient to cause b-to-a-cell reprogramming during embryonic development and in the adult. These combined in vitro and in vivo studies elucidate how cell-specific DNA modifications and transcriptional regulatory events come together to establish and maintain islet bcell identity (Papizan JB, 2011).\n\nApplications: \nSuitable for use in Immunohistochemistry. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
